The Proteas are accused of tampering with the ball in Dubai.
They were penalised five runs and the ball was changed during the Pakistan second innings today.
Faf du Plessis and Vernon Philander are the players with fingers pointed at them.
Du Plessis allegedly rubbed the ball against a zipper in his pants, while Philander allegedly picked at the ball with his fingers.
Pakistan ended day three on 132 for four in their second innings. Misbah-ul-Haq is unbeaten on 42.
Pakistan need another 287 runs to make South Africa bat again.
The Proteas were earlier dismissed for 517 in their first innings.
Graeme Smith made 234 and AB de Villiers 164.
